초록

-To study the preparation conditions of chongkukjang with strong fibrinolytic activity and characterization of fibrinolytic enzyme

-To elucidate anti-atherogenic effect and acting mechanisms of isoflavones

-To isolate and identify antidiabetic component from bean products with hypoglycemic effect

-To elucidate antidiabetic effects and mechanism of Chongkukjang in animal model of diabetes and diabetic patients

-Screening of cancer preventive components from soybean and elucidation of anti-carcinogenic mechanism of soy components

The preparation conditions of chongkukjang with strong fibrinolytic activity were determined and the enzyme with this activity was truned out to be a serine protease. The anti-atherogenic effect of isoflavones, studied in RAW 264.7 macrophages, New Zealand white rabbits, and mildly hyperlipidemic women, was mediated through its anti-inflammatory effect through NFkB down-regulation, suppressive effect of oxidative stress, and hypotriglyceridemic effect. Antidiabetic component of Chongkukjang was isolated and identified as apigenin which shows strong α-glucosidase inhibitory activity. Chongkukjang improved glycemic control and dislipidemia and increased GLUT4 gene expression in diabetic animals. Consumption of Chongkukjang for 12 weeks significantly decreased plasma glucose and improved blood lipid profile in type 2 diabetic patients. Anti-carcinogenic components present in soybean were screened and studied for possible mechanisms for differentiation-promoting and anticarcinogenic activities using cultured cancer cell model system and clinical specimen obtained from related-disease patients.

Our results support soy isoflavone is a potent anti-inflammatory, hypolipidemic agent and antioxidant that may reduce the incidence of cardiovascular disease.

Thus to eat soyfoods with protein and isoflavones or fermented soyfood such as chongkukjang with fibrinolytic activity is a wise practice in Koreans dietary life.

Dietary Chongkukjang exerts antidiabetic effects by inhibiting α-glucosidase activity and increasing GLUT4 gene expression. The results of this study could be applied to establish soybean-based recipes for diabetic patients and develop hypoglycemic functional products using Chongkukjang. A fraction shown a strong anti-carcinogenic activity was isolated from stepwise solvent fractionation and silica gel column chromatography. It induced the differentiation and apoptosis of HL 60 human promyelocytic leukemia cells very strongly. Clinical study showed that intake of soy isoflavones could reduce level of total cholesterol in prostate disease patients and improve antioxidant status in prostate cancer patients.
